چکیده

The heterogeneity among immature megakaryocytes has been examined by physical properties. cell cycle status. and responsiveness to thrombopoietic stimulatory factor. Three types of immature megakaryocytes exist that can be recognized by acetylcholinesterase staining. nuclear shape. high nucleus/cytoplasm ratio, and small size (818 ) with respect to mature megakaryocytes (>18 ii). These three acetylcholinesterase-containing cell types are distinguished by their nuclear configuration: a round, indented, and lobed nucleus. The lobed cell type was found to overlap with and enhance detection of megakaryoblasts (stage I megakaryocytes). These cells had a sedimentation velocity range of 3.5-19.0 mm hr1 and a density range of 1 .072-1 .095 9 cm . Separation of these three classes of immature megakaryocytes was achieved by equilibrium density centrifugation with modal buoyant densities of

Optimized Mouse BMDC Isolation and Culture under Endotoxin-Free Conditions

Introduction: Dendritic cells are very important in basic studies and vaccine research, but isolation and culture of these cells face challenges due to their small number in tissues. Since there is no standard method, we addressed some of the factors affecting the efficiency of dendritic cell isolation and culture from BALB/c mouse bone marrow.   Materials & Methods: Bone marrow cells isolated...

Immature Megakaryocytes in the Mouse: Morphology and quantitation by acetylcholinesterase staining.

Three types of immature megakaryocytes, detected by their morphological properties, have been characterized in bone marrow of normal C57BL/6 mice. Morphological classification of these cells was carried out by determining (1) presence and relative amount of acetylcholinesterase, (2) cell size, (3) nuclear/cytoplasm ratio, and (4) nuclear shape. The immature megakaryocytes were classified as: (A...

P-92: Assessment of Stem Cells in Adult Mouse Ovaries during Estrous Cycle

Background: The concepts of reproductive biology were changed by stating the ovarian reserve in postnatal mammalian females is replenished. The aim of this study was to investigate the expression of OCT4 in the mice ovarian tissue during different stages of mouse estrous cycle. Materials and Methods: The mice were considered as pro-estrous, estrous, met-estrous and di-estrous based on the cell ...

Podoplanin-positive periarteriolar stromal cells promote megakaryocyte growth and proplatelet formation in mice by CLEC-2.

Megakaryopoiesis is the hierarchical differentiation of hematopoietic stem cells into megakaryocytes. Differentiating megakaryocytes undergo maturation characterized by endomitosis and produce numerous platelets through proplatelet formation. C-type lectin-like receptor 2 (CLEC-2) is a podoplanin (PDPN) receptor mainly expressed on platelets and megakaryocytes. Deletion of platelet/megakaryocyt...
